Based on true historic events around 1948 during the Jeju uprising, Jiseul is about a little village which gets in trouble when the government sends army on the village's island to kill everyone as they were labelled as communists. Fearing the soldiers, the poor villagers runs towards mountains where a big cave is their hideout place. After rumbling the village, soldiers are now seeking for people everywhere else. Slowly reaching near the cave, one of the soldiers is the only hope for the villagers as he has been failing at his job of "shoot-at-sight" from the day one of his duty.

A radio host, Yoon Young-Hwa (Ha Jung-Woo), suddenly receives the call in which the caller tells him that he will blow up the Mapodaegyo Bridge. Yoon took it lightly, as a joke, but the bridge blasts off in fron of his eyes (the bridge was visible from his office), every things seems to be real and dangerous. Instead of chickening out, Yoon decides to take the caller's interview to get his career back as a TV host. But, soon he found out that it was not very wise thing to do as the caller asks for big money, a sorry-later from the president and says that he is wearing an explosive. All things seemed to be getting from bad to worse for Yoon.

Sung-Soo (Son Hyun-Joo) leading a successful life has a wife and two children. They are all happy. Though, Sung-Soo has a dark secret in his chest about his older brother. One day, he gets to know that his brother is missing who was living poor area. Out of guilt, he visits his brother's apartment where he finds some weird codes written all over his brother's apartment's walls. He also meets Joo-Hee (Moon Jung-Hee), who complains about her brother and asks him to tell his brother to stop peeping on her daughter. Joo Man-Ho (Kim Myung-Min), a marathon runner gets his leg broken in a serious injury and looses hope to become the national runner or run in the marathon for his country. Broken leg and broken dreams forces him to work at a friend's restaurant where he delivered chicken. Luckily, he succeeded to be the pacemaker of his team - the beginner of the marathon race in the team, who drops out after some distance giving the race to other members. When his coach Park Sung-Il (Ahn Sung-Ki) visits him, he offers him to become the pacemaker for the national team and run in upcoming Olympics games of 2012.
